Early postpartum ketosis in dairy cows and hepatic gene expression profiles using a bovine cDNA microarray
Abstract:
Cows had ad libitum (150% of NRC requirements) or restricted (80% of requirements) feed intake during the dry period. All cows were fed a common lactation diet after parturition. At 4 days in milk (DIM), 7 cows classified as healthy after a physical examination were fed at 50% of intake at day 4 from day 5 to signs of PK (anorexia, ataxia, or abnormal behavior) or until 14 DIM. Another group of 8 healthy cows served as controls (NK). Liver was biopsied at 10-14 DIM (PK) or 14 DIM (NK). A microarray consisting of 7,872 cDNA inserts was used for transcript profiling. Annotation was based on similarity searches using BLASTN and TBLASTX against human and mouse UniGene databases and the human genome. Cy3-and Cy5-labelled cDNA from liver and a reference standard (derived from a mixture of cattle tissues) were used for hybridizations (30 microarrays). Loess-normalized log-transformed ratios (liver/standard …
