Aislamiento y caracterización molecular de bacterias acidófilas nativas del sector minero Tundayme perteneciente a la provincia de Zamora Chinchipe


Abstract:

In the present study it was realized the isolation and characterization of native microorganisms of the mining sector Tundayme with capacity to oxidize metallic sulfides; useful for later applications in the biominería. Used medium of cultive 9k to isolate the genre Acidithiobacillus and Leptospirillum with values of pH (6,5, 1.2, 2.4, 1,8) and temperatures (20 and 30°C). Were obtained 48 initial tests of which it selected the 6 better cultives (M1, M3, M5, M7, M9, M11), later determined their oxidating capacities through quantification of the consumption of Fe2+, production of Fe3+ and sulfates. Finally it was managed to obtain gram-negative microorganisms, that present the region 16S rADN with sizes of band approximated to 1500pb; of equal way they presented good capacity to oxidize iron and sulfur; the optimal conditions in culture medium for isolated 1 was pH 1.8 and 30 °C which had volumetric productivity (Qp) of 0.530 g/Lday ferric ion (M7), for isolated 2 were pH 6.5 and 30 °C with volumetric productivity (Qp) was 0.116 g/Lday sulfate (M9) and for isolated 3 were pH 1.2 and 30 °C with volumetric productivity (Qp) of 0.340 g/L ion ferric.
