Estrategias para mejoramiento de expresión de proteínas en el cloroplasto de Chlamydomonasreindhardtii
Abstract:
World population will demand 70% more food and 50% more energy and water by 2050, promising green alternatives, such as microalgae can be engineered to produce biomass for animal and human food, high valuable products (eg therapeutic proteins, hormones, antibodies), industrial enzymes and biofuels. However, recombinant protein production is poorly developed, and no efficient system is known for transgene expression. Combinatorial approach, where different cis-acting elements are tested, showed to be the strategy to improve foreign protein production in Chlamydomonas reinhardtii chloroplast. The endogenous Prrn promoter and four 5’-UTRs belonging to the major capsid protein gene of cyanophages P-SSP7, Pf-WMP4, P-SSM2 and P-SSM4 were fused to the reporter gfp gene.
