First report of apple mosaic virus infecting Rosa spp. in Pichincha province, Ecuador


Abstract:

Rose is a member of the family Rosaceae and is widely cultivated in different parts of the world. Between July and August 2015, plants exhibiting mosaic and mottling symptoms, bright yellow ring spots and line patterns were observed in rose plants from Pichincha province, Ecuador. Based on these symptoms, the presence of apple mosaic virus (ApMV) from the genus Ilarvirus in the family Bromoviridae was suspected. Symptomatic leaf samples (n= 15) were screened via DASELISA (Agdia, USA), of which, four were found to give a positive reaction. Total RNA was isolated from the four positive leaf samples using PureLink™ Plant RNA Reagent (Invitrogen, USA) and an ApMV coat protein gene fragment (~ 500 bp) was amplified by RT-PCR (Li et al. 2009).
