Generation of artificial seeds of blackberry (Rubus glaucus Benth) from organogenesis
Abstract:
This research established artificial seeds of blackberry (Rubus glaucus Benth) from direct organogenesis. The vegetal tissue used for this investigation were apical and nodal segments t. The disinfection o was done with different concentrations of sodium hypochlorite (0.5, 1 and 1.5%) and immersion times (10 and 15 minutes). It was obtained the less value of pollution and tissue death in the treatment that consist on 1.5% Sodium Hypochlorite and 10 min of immersion time. Shoots were induced from nodal and apical segments on MS basal medium containing various concentrations of TDZ (0.25, 0.5 0.75 and 1 mg. L-1). medium which yielded the best response in the treatment 0.5 mg. L-1 TDZ. In the multiplication stage the explants were induced on MS basal medium containing various concentrations of AIA (0.5, 0.75 and 1 mg. L-1) combined with BAP (2 and 3 mg. L-1). The most effective combination for development of the largest number of internodes and longest plants was 2 mg. L-1 of BAP and 0.75 mg. L-1 of AIA. Finally, obtained the optimal explants, they were encapsulated in sodium alginate (3%) and calcium chloride (1%) dissolved on MS combined with different concentrations of brassinolide (0.1, 0.5 and 1 mg. L-1) and characoal (0 and 1 g L-1) obtaining the best response in the treatment BRA 1 mg. L-1 and 1 g L-1 activated charcoal, allowing germination in vitro of blacberry.
