Identification of archaea community of the Papallacta geothermal source, located in Napo province–Ecuador, through new generation sequencing
Abstract:
The geothermal sources are an extreme environment allocated around the world with unique physicochemical characteristics. Culture-independent molecular studies have evidenced that moderate temperature geothermal sources (50-70 C) have a greater diversity of archaea. The objective of this research was to identify the archaea community in water and sediment samples from the Papallacta geothermal source, through an Illumina MiSeq Platform to sequence 16S rRNA gene using the primers Arch349F/Arch806F. In addition, the physicochemical characterization of the samples was carried out. In a first sampling, 38311 OTUs were identified in water and also 104135 OTUs in sediment. The presence of OTUs in water were mostly classified in Thaumarchaeota (74.68%), while in the sediment it was Euryarchaeota (77.55%).
