Induction of micronucleated reticulocytes in peripheral blood of mice receiving a suspension of Chuquiraga jussieui J.F. Gmel
Abstract:
Introduction: The plant species that grow in the Andes mountains range have developed unique characteristics only found in these ecosystems of the American continent, characterized by their abundant and variegated flora. Particularly in Ecuador a number of different species, among them, Chuquiraga jussieui JF Gmel, are known to have been traditionally used to treat various conditions, though thorough studies are not available about the safety of the extracts obtained from them. Objective: Determinate whether a C. jussieui suspension induces the formation of micronucleated reticulocytes in the peripheral blood of mice as an indicator of clastogenicity and aneugenicity. Methods: Ultravital staining with acridine orange was used to evaluate the potential of a C. jussieui suspension to produce micronucleated reticulocytes in the peripheral blood of mice of the NMRI line. The suspension was administered orally in 3 dose at a rate of 10 mL/kg body weight. The frequency of micronucleated reticulocytes was evaluated at 48 and 72 h after administration, and the frequency of such alterations was observed in 2 000 cells per animal at each evaluation time. Results: Significant statistical differences were not found for p< 0,05 in the number of micronucleated reticulocytes between the treatment groups with respect to the vehicle control. However, significant differences were found with respect to the genotoxicity pattern control. Conclusions: The study substance did not show any clastogenicity or aneugenicity in these experimental conditions.
