Influencia de la criopreservación en perlita y pda sobre la viabilidad de basidio y ascomicetos del cepario hutpl f.
Abstract:
Fungi are organisms with high levels of bioprospecting, however, there are limitations to their correct study, such as: difficulty of collection, identification and in vitro preservation. For this reason, this research work, has the following purpose, the implementation and evaluation of methods for the conservation of basidiomycete fungal strains (ie, Armillaria sp., Gymnopus sp., Lentinus sp., and Pleurotus sp.) and Ascomycetes (ie, Pestalotiopsis sp.) in a long term. Two conservation methods were assement, the first one is the Perlite Homolka Protocol (-80ºC) Modified and the second one the PDA Medium Protocol. On these mycelium of the different strains were inoculated and monitored after monthly reactivation during a year. The data obtained indicate a reactivation of 100% in the PDA medium for the strains Gymnopus sp., Lentinus sp., and Pestalotiopsis sp. however, the Homolka Perlite modified method (-80ºC) allowed to keep the macro and microscopic characteristics for all the strains, for this reson we suggest this method and medium as the most generalist for the evaluated species. These data indicate that the strains react differently according to the conservation method.
