Micropropagation of pollizo V4 - CIDA selection
Abstract:
In this study, the optimal conditions for the establishment, proliferation and rooting stages of the Pollizo V4 CIDA rootstock selection are determined. The establishment stage for the explants was carried out on Murasighe and Skoog, MS (1976) medium, achieving 75% sprouting. In the proliferation stage, different concentrations of BAP (0, 0.2, 0.5, 1, 1.5, 2, 2.5 and 3 mg/l) and IBA (0, 0.1, 0.2, 0.3, 0.4 mg/l) were studied, again using the MS culture media, with 7 g/l of Bacto Agar Difco and 30 g/l of sucrose. The best results, with rate of proliferation of 7.9, were obtained with 1 mg/l of BAP + 0.1 mg/l of IBA. We also compared three culture media: MS, Loyd and McCown, WPM (1980) and Driver and Kuniyuki, DKW (1984), each with the best growth regulator concentrations determined previously. Significant differences in the proliferation results were not observed. With respect to the rooting stage, a trial with different IBA concentrations was made: 0, 0.2, 0.5, 1, 1.5, 2, 2.5 and 3 mg/l, were compared in the MS culture media, with 7 g/l of BactoAgar Difco and 30 g/l of sucrose; 61,11% rooting was achieved with 3 mg/l of IBA. With the purpose of increasing the percentage of rooting, another trial was carried out by dipping the explant base for 10 seconds in concentrated solutions of IBA (0, 250, 500, 750 and 1000 mg/l); this produced 100% rooting explants with all the auxin treatments, which shows the effectiveness of this rooting induction method.
