New approach to simultaneously identify mitochondrial DNA haplogroups by multiplex PCR-RFLPs and capillary electrophoresis


Abstract:

Mitochondrial DNA (mtDNA) genotyping provides a valuable tool for forensic studies and population genetics. Nevertheless, current methodologies for mtDNA genotyping are laborious and expensive. Here, we describe a detailed new multiplex PCR-RFLPs and capillary electrophoresis approach to simultaneously identify six mtDNA haplogroups: A, B, C, D, H & L. 38 samples previously typed by sequencing of the complete mtDNA control region (CR) were analyzed by this methodology. Haplogroups determined by RFLP-PCR for all samples corresponded with the results obtained by sequencing. The results show that this approach effectively allows the simultaneous determination of mtDNA haplogroups commonly present in Latin American populations in a relatively fast and economical way. This methodology offers a good resolution and sensitive detection procedure to assess mtDNA ancestry.
