Pituitary hormonal response to the orthopramides clebopride, bromopride, metoclopramide, and sulpiride
Abstract:
The orthopramides metoclopramide, sulpiride, and clebopride have been shown to rapidly elevate serum prolactin (PRL) after a single oral dose or a single parenteral injection. However, we are not aware of any study comparing the PRL response to these orthopramides as well as the effect of bromopride on PRL secretion. On the other hand, the incidence of reproductive side effects such as anovulation, amenorrhea, and/or galactorrhea is variable for these drugs. In this study, we have investigated whether these differences may be due to different effects upon the dopamine receptors of the tuberoinfundibular axis by comparing their ability to induce PRL release at clinically comparable doses. In addition, luteinizing hormone (LH) follicle-stimulating hormone (FSH), thyroid-stimulating hormone (TSH) and growth hormone (GH) serum levels were determined.
