Recombinant expression of active Penicillium minioluteum endodextranase in alternative hosts


Abstract:

The dextranase gene from the fungus Penicillium minioluteum was expressed in Escherichia coli. The recombinant endodextranase represented 1.5% of the total soluble protein. Crude recombinant enzyme preparations displayed a product profile of dextran hydrolysis as well as heat stability properties similar to those of the naturally glycosylated enzyme. Transient expression of the fungal dextranase in bombarded tobacco cells yielded dextran hydrolytic activity.
