Validation of an analysis method through high performance liquid chromatography for determination of quercetin in allium shoenoprasum regel &tiling (Chives)


Abstract:

Introduction: Allium genus species such as onions, garlic, leeks and chives are known for being widely used in most traditional pharmacopeia worldwide. Among these genus species, Allium schoenoprasum L (chives) has been barely studied, especially from the pharmacognostic point of view. In a study using high performance liquid chromatography and mass spectrometry conducted by Ulla (2001), there were found quercetin, kaempferol and isorhamnetin weighing 3, 12 and 5 mg/100 g in the species extracts. Objective:To validate an analytical method through high performance liquid chromatography (HPLC) to quantify quercetin in Allium schoenoprasum extract. Methods: for the study, the KNAVER liquid chromatograph was used under the following working conditions: mobile phase phosphate buffer solution pH=2.4: acetonitrile (70:30); oven temperature set at 25 °C; detector called KNAVER by diode array, 370 nm (200-400); flow at 0.9mL/min; 20μL injection volume; Uptisphere 5 708 965 0DB column (RP-18). Results: The validated method was linear, precise, accurate and specific. Conclusions: This method is simple and reliable and can be used for quality control of Allium schoenoprasumL(chive) and their preparations.
