Carrier Molecules Displaying Dual Haptenic Presentation for in Vivo Testing to Determine IgE Antibody in Patients Allergic to Betalactams

Abstract:

Methods Well-defined hapten-carrier conjugates were synthesized by haptenization of 4 th generation PolyAMidoAMine dendrimers with either one kind of penicillin (benzylpenicillin and amoxicillin) or both penicillins. The different conjugates were immunologically tested by RAST inhibition using sera from 4 controls and 7 patients allergic to penicillins, both selective and cross-reactive responder. Results At the maximum conjugate concentration, all sera allergic to benzylpenicillin showed 80% of RAST inhibition to both benzylpenicilloyl-dendrimer and mixed-dendrimer, and negative to amoxicilloyl-dendrimer. All sera allergic to amoxicillin showed 90% of inhibition to both amoxicilloyl-dendrimer and mixed-dendrimer, with no inhibition to benzylpenicilloyl-dendrimer.
