Chemical composition and antimicrobial activity of ethanol extract of aerial parts of Minthostachys mollis griseb


Abstract:

Introduction: Most studies about Minthostachys mollis Griseb are related with the content of essential oils and their antimicrobial potential; however, scarcely exploration has been made in such aspects in the extracts of the aerial parts of the plant. Objective: To evaluate the chemical composition and in vitro antibacterial activity against Staphylococcus aureus of the ethanolic extract of M. mollis. Methods: Some work was carried out with the aerial parts of the species in the flowering period. A phytochemical screening was performed on three extracts of increasing polarity. The ethanolic extract obtained by maceration was evaluated by applying some physical-chemical parameters (pH, refractive index, relative density, total solids). After depigmenting the extract, it was analyzed by gas chromatography adapted to mass spectrometry. The mass spectrum was performed by direct infusion. In a collateral way, the Folin-Ciocalteu trial was performed to determine the preliminary antimicrobial activity against Staphylococcus aureus. Results: Phytochemical screening suggested the presence of oils and fats, triterpenes - steroids, phenolic compounds, reducing sugars, saponins, amino acids and flavonoids. The characterization of the depigmented extracts by GC-MS demonstrated the presence of sugars, amino acids (alanine, leucine, isoleucine, proline, phenylalanine), phenolic compounds (caffeic acid, isomers of caffeic acid, catecholactate) and a triterpene (majority compound). Mass spectrometry analysis by direct infusion allowed the identification of rutin. The Folin-Ciocalteu trial demonstrated the existence of phenolic compounds and metabolites with reducing capacity, and the extract showed sensitivity to Staphylococcus aureus. Conclusions: The ethanolic extract of the vegetable drug studied could be used due to its chemical composition and antibacterial activity as a complementary natural medicine in disorders caused by S. aureus.
