Abstract:
Bacterial or viral infections trigger flares of autoimmunity, but the mechanisms are poorly understood. Toll-like receptors (TLR) recognize multiple pathogen-related antigens whereas TLR9 specifically interacts with unmethylated CpG-DNA. As TLR9 activation with exogenous or endogenous CpG-DNA may contribute to disease activity of systemic lupus erythematosus, we examined the effects of CpG-oligodeoxynucleotides (ODN) or DNA derived from E. coli on the course of autoimmunity in MRLlpr/lpr mice, a model of lupus nephritis. Messenger RNA and protein for TLR9 were present in kidneys of MRLlpr/lpr mice and localized to glomeruli, tubulointerstitium and perivascular areas.
