Evaluación de las variables altitud y composición del suelo sobre la actividad antibacteriana y antifúngica de extractos lipofílicos de hypericum laricifolium juss por fingerprint cromatográfico
Abstract:
Hypericum laricifolium Juss is used for the treatment of microbial infections. The secondary metabolic profile and hence the biological activity could be influenced by several environmental variables. The aim of this work was to evaluate the effect of altitude and soil compositions on the antibacterial and antifungal activity of lipophilic extracts of Hypericum laricifolium, by evaluating 29 individuals and their soil samples obtained in different environmental conditions and to get a pbkp_redictive model of their effect using fingerprint chromatography by HPLC. Soil samples were analyzed for pH, electrical conductivity, organic matter, nitrogen, phosphorus and potassium. The extracts were obtained by supercritical fluid extraction. For molecular identification, the loci ITS1, ITS2 and rbcL were sequenced. The identity for ITS1 and ITS2 was 100% and 99.7% respectively, rbcL exhibited genus identity (98,5%). The antibacterial activity was performed by micro-dilution technique against MRSA. A high activity has been detected. Using partial least squares (PLS) a correlation was established between antibacterial activity and chromatographic profiles at 254, 280 and 360 nm. The model was constructed using the individuals with their replicas, with 7501 X variables (responses at different retention times), variable y (IC50 of each individual). The model was evaluated using R2 correlation parameters, the model at 254 nm pbkp_redicted better the antibacterial activity (R2=0.92).
