Protein extraction from intercellular space of banana leaves, cv.‘Grande naine’(Musa AAA)
Abstract:
The extraction and characterization of proteins involved in plant pathogenesis process, has been an important tool for pathosystem study. The study of secreted proteins to intercellular space of leaves in response to biotic and abiotic factors constitutes an important step to reach this purpose. Intercellular proteins, from healthy and infected leaves tissue of banana plants Musa AAA (subgroup Cavendish) cultivar ‘Grande naine’ at different stages of development of the Black Sigatoka disease were extracted. Leaves fragments of 2 cm2 were infiltrated at vacuum, centrifuged and 80% (w/v) of ammonium sulphate for protein precipitation was added to the collected intercellular fluid , which were analyzed in a denaturant polyacrilamide gel at 12%. The average protein concentration obtained was 0.43 mg.ml-1 for healthy and 0.83 until 1.0 mg.ml-1 for infected samples from intercellular space. The results are the first step in Musa-Mycosphaerella fijiensis pathosystem study, allowing to know about the proteins involved in the interaction, as an essential aspect to take into account for the development of future strategies of resistance to pathogen.Key words: denatured electrophoresis, protein purification, salt precipitation
